| چکیده [English] | ||
| Alternative splicing (AS) is a post-transcriptional regulatory mechanism that plays an important role in regulating plant stress responses by increasing transcript and proteome diversity. In this study, the effects of 3-day drought, 6-day drought, and 1-day water recovery treatments on alternative splicing and gene expression at the transcriptional level in maize seedlings were investigated. Transcriptomic data analysis using the bioinformatics tool CLC Genomics Workbench and Python scripts identified transcripts exhibiting alternative splicing events in the aforementioned treatments. Among the identified events, alternative splicing patterns involving A5′SS (Alternative 5′splice site) and A3′SS (Alternative 3′splice site) positions were rarely observed, whereas other splicing patterns were the most frequently detected events. Functional enrichment of alternative splicing-related genes using STRING and KEGG databases confirmed their pivotal role in cellular processes, metabolic regulation, especially the metabolism of selenium-containing compounds and the degradation of branched-chain amino acids. Functional analysis of differentially expressed alternatively spliced genes (DE-ASGs) revealed their roles in lignin biosynthesis, lipid transport and metabolism, plant defense responses, growth and development, and hormonal signaling, which could be considered as major targets in breeding programs. In addition, predicted miRNA-mRNA interactions using the psRNATarget web server revealed the microRNAs zma-miR166j-5p, zma-miR399f-5p, zma-miR2275a-3p, and zma-miR2275d-3p as candidate regulators of alternatively spliced transcripts. These findings highlight the importance of alternative splicing as an approach to enhance transcriptional flexibility and reprogram plant molecular pathways and thereby increasing drought stress resistance. | ||
