بررسی تاثیر القای پلیپلوئیدی در ماهی قزلآلای رنگینکمان (Oncorhynchus mykiss) بر رشد، بقا و گلبولهای قرمز | ||

در این پژوهش، ماهیان دیپلوئید، تریپلوئید و تتراپلوئید قزلآلای رنگینکمان از نظر رشد، بقا و ویژگیهای گلبول قرمز مورد مقایسه قرار گرفتند. برای تولید ماهیان تریپلوئید، تخمهای لقاح یافته تحت شوک گرمایی 5/26 درجه سانتیگراد در قالب سه تیمار: 1- دوبار شوک یک دقیقهای در 15 و 30 دقیقه بعد از لقاح، 2- یکبار شوک 15 دقیقهای در 15 دقیقه بعد از لقاح و 3- یکبار شوک 12 دقیقهای در 20 دقیقه بعد از لقاح، قرار گرفتند. برای تولید ماهیان تتراپلوئید، تخمهای لقاح یافته تحت شوک گرمایی 28 درجه سانتیگراد در قالب سه تیمار: 1- شوک 10 دقیقهای در 59 ساعت-درجه بعد از لقاح، 2- شوک 10 دقیقهای در 66 ساعت-درجه بعد از لقاح و 3- شوک 10 دقیقهای در 72 ساعت-درجه بعد از لقاح، قرار گرفتند. در تریپلوئیدسازی، شوک گرمای 5/26 درجه سانتیگراد در 20 دقیقه بعد از لقاح بهمدت 12 دقیقه با درصد تفریخ 6/66 درصد و نرخ تریپلوئیدسازی 87 درصد بهتر از سایر گروهها بود. در تتراپلوئیدسازی، شوک گرمای 28 درجه سانتیگراد در 59 ساعت-درجه بعد از لقاح بهمدت 10 دقیقه با درصد تفریخ 9/54 درصد و نرخ تتراپلوئیدسازی 94/7 درصد نسبت به سایر تیمارها مناسبتر بود. ماهیان تریپلوئید از نظر رشد با دیپلوئیدها اختلاف معنیداری نداشتند اما رشد ماهیان تتراپلوئید بهطور معنیداری کمتر از دیپلوئیدها بود. بهطورکلی، در این مطالعه علاوه بر بهینهسازی روشهای تریپلوئید و تتراپلوئیدسازی، استفاده از طول، عرض و مساحت سلولی گلبولهای قرمز، بهعنوان یک روش کاربردی و کم هزینه برای تفکیک ماهیان دیپلوئید، تریپلوئید و تتراپلوئید قزلآلای رنگینکمان مورد تائید قرار گرفت. | ||

In this study, rainbow trout diploid, triploid, and tetraploid were comparatively investigated regarding the growth performance, survival rate, and erythrocyte characteristics. For the triploid production, the fertilized eggs received 26.5 ℃ thermal shock under three treatments: 1. double 1-min shock after 15 and 30 min of the fertilization, 2. one 15-min shock after 15 min of the fertilization, and 3. one 12-min shock after 20 min of the fertilization. Tetraploidy was induced by application of 28 ℃ thermal shock under three treatments: 1. 10-min shock after 59 degree-hours of the fertilization, 2. 10-min shock after 66 degree-hours of the fertilization, and 3. 10-min shock after 72 degree-hours of the fertilization. The best triploidization results, 66.6% survival rate and 87% triploidization rate, achieved by application of 12-min shock after 20 min of the fertilization. The best tetraploidization results, 54.9% survival rate and 7.94% triploidization rate, achieved by application of 10-min shock after 59 degree-hours of the fertilization. There were no significant differences between the triploid and diploid fish, wheras the tetraploids showed significantly lower growth rate than the diploids. In conclusion, in this study, triploid and tetraploid production of rainbow trout were improved, and application of erythrocyte cell size as a reliable and efficient polyploidy detection method was emphasized. | ||
