بررسی میزان تولید کاتچین در عصاره برگ و ریشه مویین تراریخته گیاه Hibiscus sabdariffa و تأثیر آن بر سلولهای سرطانی کبد HepG2 | ||

چکیده | ||
در این پژوهش به منظور ارزیابی ترکیبات مؤثر موجود در عصاره گیاه چای ترش (Hibiscus sabdariffa) و تأثیر آن بر رشد سلولهای سرطانی رده HepG2 مقایسهای بین میزان متابولیتهای ثانویه موجود در عصاره بافت برگ گیاه و عصاره ریشه مویین تراریخته آن انجام گرفت که در آن میزان آنتوسیانینها و فلاونوییدها و فنل کل در هر دو اندازهگیری شد. در طی انجام سنجش پلیفنلها با دستگاه HPLC میزان غلظت فلاونویید کاتچین در عصاره ریشه مویینه تراریخته به میزان 8/22 میلیگرم بر گرم ماده خشک تعیین شد که چندین برابر مقدار 9/0 میلیگرم بر گرم در نمونه عصاره برگی چای ترش ماده خشک بود و این نشاندهنده افزایش معنیدار در غلظت این متابولیت ثانویه مؤثر نسبت به سایر متابولیتهای ثانویه در طی فرایند تولید ریشه مویین تراریخته میباشد. سپس عصاره نمونههای برگی وریشه مویین در غلظتهای مختلف صفر، 25، 50، 100، 200 و 400 میلیگرم بر میلیلیتر به سلولهای سرطانی کبد HepG2 در آزمایشگاه اضافه شدند ودرصد زندهماندن سلولها بهروش MTT محاسبه شد. میزان زندهماندن سلولها در نمونههای تحت تیمار با عصاره ریشه مویین تراریخت کمتر از نمونه چای ترش بود که به احتمال زیاد بهدلیل وجود غلظت بالاتری از کاتچین در نمونه ریشه مویین تراریخته میباشد و بهترین غلظت عصارههای نمونه چای ترش و ریشه مویین تراریخته که بالاترین درصد آپاپتوز (مرگ سلولی) سلولها را نشان داد غلظت 100 میلیگرم بر میلیلیتر بود. | ||

In this research, a comparison was made between the concentration of secondary metabolites of Hibiscus sabdariffa content in its leaves and induced transgenic hairy roots. The levels of anthocyanins and flavonoids and total phenol were measured in both samples. In the next step, polyphenols were measured by HPLC. The concentration of catechin flavonoids in the transgenic hairy root extract was 22.8 mg/g dry matter which was several times higher than that of 0.9 mg/g in leaf extract of dry matter. The obtained results clearly demonstrated that, the rate of concentration of secondary metabolites during the process of induction of transgenic hairy root was increased. Then, the extracts of leaf and transgenic hairy root samples at various concentrations of 0, 25, 50, 50, 100, 200, 400 mg / ml were added to HepG2 liver cancer cell lines and the percentage of cell survival was calculated by MTT method. The cell survival rate in the sample treated with transgenic hairy roots samples was less than the leaf extract samples, which was most likely due to the higher concentration of Catechin in the transgenic hairy roots extract. The best concentrations of extracts of transgenic hairy root extract showed that, the highest percentage of cell apoptosis was 100 mg/ ml. | ||
